When this question arises in your head, if to let go or try a bit more, you know its time to let go then and there itself.
When stuck with a dilemma, I sometimes pen down the actual pros and cons. You could also design a point system with an end goal in mind for each one. The physical representation can sometimes make the issue seem smaller than it really is or help you make a decision as you're jotting everything down.
It depends. Is it a person you want to let go of or is it a thing/dream/goals you wanna let go of?

If it's a person, it's easy. Once they say no, you stop. Maybe for your own happiness you can try twice but honestly once is enough. You cannot make someone love you. You cannot keep on trying harder and harder to mend relationships. Just because you want them, just because you think you are a good person, it doesnt mean they want you too. They have free will. And their will to be away from you triumphs your will to be with them.

But if this person is your child, or your pet, or your parents who are too old and are completely dependent on you then you try more than once. You try multiple times till it wears you out.

And if its a thing, like a goal or something, you try until it starts affecting your mental health.

I have 5 years to UPSC exam, a childhood dream. I stopped because it completely wrecked my mental health. Because it made me feel super underconfident after every rejection. If it was person, i wouldn't have kept coming back every year thinking that things would change.
